Foster's Home for Imaginary Friends is about an 8-year-old boy, Mac, who is forced to take his imaginary friend Blooregard Q. Kazoo ("Bloo" for short) to a foster home for imaginary friends called, yes, Foster's Home for Imaginary friends. It is a facility run by a nice old woman (named Madame Foster) for all abandoned imaginary Friends. Though Mr. Herriman, the strict taskmaster of the home, hates it, Mac makes a deal with Madame Foster that Bloo won't be adopted as long as he visits all the imaginary friends frequently.

There are an assortment of imaginary friends other than Bloo that Mac interacts with; again, there's Mr. Herriman (who, as it turns out, is Madame Foster's imaginary friend); Eduardo, who looks like he's from Where the Wild Things Are—he may look scary, but is scared of almost everything; Wilt, a tall, one armed monster that's over-polite; and Coco, a bird that can lay eggs with objects in them and only says "Coco".

Mac also interacts with human characters, including Madame Foster; Frankie, Foster's granddaughter who cleans up the place; and Terrance, Mac's evil older brother.

The episodes almost always revolve around one of the imaginary friends or Foster's; and the episode titles are often puns (e.g., "House of Bloo's").

The show was nominated for an Emmy a number of times. It won a handful of special juried Emmys, and won the big Emmy for Outstanding Animated Program (More Than One Hour) for the special longform episode "Destination Imagination."

Creator Craig McCracken maintained ties of sorts with his previous creation The Powerpuff Girls in this series--Frankie's T-shirt shows color-coordinated silhouettes of the Powerpuffs, Mojo Jojo appears in the debut movie, and in a Halloween episode, three members of the home are dressed as the girls (Frankie as Blossom).
